Stock Solutions for RNA Work

10X MOPS Buffer

3-(N-morpholino)-propane sulfonic acid40 g per Liter
Sodium Acetate2 g per Liter
500 mM EDTA (autoclaved)20 mL per Liter
Dissolve components in 980 mL deionized water, pH to 7.0 with NaOH (takes several pasteur pipettes worth of 10 N). QS to 1 Liter with deionized water. Filter. Do not autoclave MOPS. Wear gloves when preparing. Wrap bottle in aluminum foil. Store at 4C.
 

RNA Sample Buffer (per mL)

Formamide562 uL
Formaldehyde (37%)187 uL
10x MOPS112 uL
ddH2O128 uL
Ethidium Bromide (10 mg/mL)11 uL
Make about 10 mL and aliquot 500 uL portions into eppendorfs. Store at -80C. Treat as 2X. There is special store of EtBr for use with RNA.
 

RNA Formaldehyde/Agarose Gel

Rnase free agarose1.2 g
ddH2O85 mL
10X MOPS10 mL
Formaldehyde (37%)5 mL
Microwave agarose and water. Cool to 60C, then add MOPS and formaldehyde in the hood. Pour gel.
 

DEPC Water (per mL)

  • Add 1 mL diethyl pyrocarbonate to 1 Liter of distilled water.
  • Shake vigorously to get the DEPC into solution.
  • Allow solution to stand for 5-6 hours
  • Autoclave (liquid, 20 minutes)
